Y-Biologics’ PD-L1/CD3 Bispecific Antibody
Y-Biologics has signed a government-wide life-cycle new drug development project agreement with the Government-wide New Drug Development Project Group to develop ACE-05, an anti-cancer dual antibody with increased T cell activity. According to the agreement, Y-Biologics will be carrying out research on the efficacy and mechanism of ACE-05, an anticancer drug candidate, with the support of the project group. ACE-05 is a PD-L1/CD3 bispecific antibody that inhibits immune checkpoint molecules and binds to T cells and cancer cells simultaneously. ACE-05 is a bispecific antibody candidate based on ALiCE (Antibody-Like Cell Engager), the company’s new bispecific T cell engager platform. It has shown to exhibit powerful efficacy even at low doses compared to the existing PD-L1 antibodies in tumor models. The ALiCE platform developed by Y-Biologics recognizes the CD3 of T cells and a target antigen on the surface of cancer cells at the same time. Compared to Amgen’s BiTE, it was designed to bind more strongly to cancer cells by optimizing the cancer cell-immune cell distance. The results of animal testing showed that the toxicity was lower, and it demonstrated substantially improved stability in the bloodstream, with more than double the molecular weight of BiTE (50 kDa), as well as enhanced productivity and efficacy. Although the PD-1/PD-L1 type immune checkpoint inhibitors currently on the market exhibit excellent efficacy, only 20-30% of cancer patients respond to them. On the other hand, CAR T cell therapy has a high response rate of over 80% in blood cancer patients, but its use is limited, as it must be tailored to each patient and it is expensive. Y-Biologics explained, “It is expected to provide an alternative to CAR-T cell therapy, which is costly, by addressing the shortcomings of the existing immunotherapy with an increased response rate and activating immune cells through antibodies." 대전 유망 바이오기업 투자포럼 결성..28일 첫 행사

Young Woo Park, CEO of Y-Biologics, a bio startup that develops antibody drugs, revealed his aspirations to localize immune checkpoint inhibitors in an interview with Maeil Business Newspaper. What is an immune checkpoint inhibitor that makes “localization” so important? When the immune cells in our body locate cancer cells, they immediately recognize and attack them. Problems arise, however, when the immune cells are too weak to effectively fight against cancer cells or if they attack normal cells. This is why immune cells have a sort of a brake device to regulate their activity, and this is called an “immune checkpoint.” The problem is that if the brake is applied excessively, the immune cells cannot function properly or kill cancer cells in a timely manner. In recent years, immune checkpoint inhibitors that can prevent such brake malfunctions have been a hot topic in the fight against cancer in the pharmaceutical industry. Because this class of drugs uses the patient’s own immune system, it does not lead to any adverse reactions like the existing chemotherapy drugs, and if the patient responds to the drug, it can increase the survival period by up to 5 years. “Recently, most pharmaceutical companies have been testing to see whether the effectiveness of anticancer drugs gets doubled when used together with immune checkpoint inhibitors,” said CEO Park who then added, “Foreign immune checkpoint inhibitors, however, are expensive, and their developers have no reason to conduct joint research with a Korean pharmaceutical company since there are companies lining up to request clinical trials on combination therapy.” He then explained that localization is essential to meet the clinical demand of domestic pharmaceutical companies and to reduce the financial burden on the national health insurance scheme by providing alternatives to the costly foreign-made immune checkpoint inhibitors. There is an urgent need to develop inhibitors targeting PD-1 and PD-L1, in particular. PD-L1 found on cancer cells weakens immune cells by triggering PD-1, which is a type of a brake for immune cells. PD-L1 also disrupts the immune system by disguising cancer cells as immune cells. CEO Park said, “Cancer cells with PD-L1 can evade the attacks from immune cells by pretending to be immune cells. [...] Since PD-1 and PD-L1 are commonly found on cancer cells, it will be possible to conquer various types of cancer by developing an inhibitor targeting them.” Antibody drugs that bind to PD-1 and PD-L1 to block their functions and normalize the immune system are being developed one after another worldwide, and the prime examples of these are Opdivo and Keytruda, which have recently been registered for national health insurance coverage in Korea. CEO Park further explained, “The global immuno-oncology drug market is expected to grow to KRW 30 trillion in size by 2022, and Opdivo and Keytruda have 80% share of the entire market. [...] The financial burden on the state will surge due to the registration of these two products for national health insurance coverage, but the patents for Opdivo and Keytruda will both expire in 2030, so it’s not even possible to release biosimilars until then.” In order to get into this niche market, Y-Biologics is currently developing immune checkpoint inhibitors with the target launch year set as 2025 prior to the patent expiry date. CEO Park said, “There are few companies in Korea that possess PD-1 and PD-L1 antibodies. The goal is to kick off a Phase 1 clinical trial next year after entering the preclinical phase (animal testing) for an immune checkpoint inhibitor candidate this year. [...] We have been delivered research findings that it is more effective than the existing immune checkpoint inhibitors from a pharmaceutical company reviewing the joint study, so it will be an excellent alternative once it is released.” Y-Biologics has already discovered PD-1 and PD-L1 antibodies using its own antibody library and high-speed antibody search system. This was possible because the company has a database on 100 billion human antibodies. CEO Park explained, “We are a R&D-oriented company that introduces new antibody drug pipelines and offers research services based on our core technologies. [...] 44 out of the 52 executives and employees at our company are R&D personnel.” The company specializes in research to find candidate substances, in particular. The company even boasts its own human antibody library. “It is like a treasure trove of antibodies exhibiting great diversity, making it possible to quickly search for antibodies needed to treat specific diseases, and it differentiates us from companies that outsources the search work or obtains antibodies from foreign companies,” said CEO Park. Each antibody has a site with a unique structure and shape for recognizing and binding to a specific antigen. It is similar to the way a key can fit only in its own keyhole to lock or unlock it. Y-Biologics’ high-speed antibody search system finds the best candidate by identifying the antibody that binds best to the antigen in question and efficiently removes the disease-causing substance. The company is also developing new antibody therapeutics based on ALiCE, a bispecific antibody technology. It is a new drug that has overcome the shortcomings of CAR-T cell therapy, which has recently been in the spotlight. CAR-T therapy designed to attack only cancer cells by manipulating a group of immune cells called T cells has been noted for its high response rate, but the downside is that it is difficult to mass produce and its effects are relatively short-lasting. “The ALiCE platform can recognize antigens better than the existing antibodies, and it has a stronger binding affinity to antigens and stays in the blood longer,” said CEO Park who then explained that “one antibody binds to a cancer cell and the other activates a T cell.” He then revealed that there are “plans to proceed with animal testing as soon as possible.” [By Yun Jin Kim]

[MK바이오스타 기업 탐방] 박영우 와이바이오로직스 대표
항체 의약품을 개발하는 바이오 벤처 와이바이오로직스의 박영우 대표(사진)는 매일경제와 인터뷰하면서 이 같은 포부를 밝혔다. 면역관문억제제가 무엇이길래 이토록 `국산화`의 필요성을 강조하는 것일까. 우리 몸에 있는 면역세포는 암세포를 발견하면 곧장 적군으로 인식해 공격한다. 그런데 면역세포가 비실비실해서 암세포와 잘 싸우지 못해도 몸이 고장나고, 엉뚱한 세포를 시도 때도 없이 공격해도 골칫거리다.이에 따라 면역세포는 강약 조절을 위한 일종의 브레이크 장치를 두는데, 그게 바로 `면역관문`이다. 문제는 브레이크가 자꾸 걸리면 면역세포 기능이 약해지거나 제때 암세포를 죽이지 못한다는 것이다. 이에 최근 제약업계에서는 암 퇴치를 위해 브레이크의 오작동을 막는 면역관문억제제가 화두로 떠오르고 있다. 환자 본인의 면역을 이용하기 때문에 기존 항암제처럼 면역거부 반응을 일으키지도 않고, 약이 잘 들으면 최대 5년까지도 암 환자 생존기간을 늘려주기 때문이다. 박영우 대표는 "최근에는 대부분 제약사들이 개방 중인 항암제를 면역관문억제제와 함께 썼을 때 (병용) 효과가 배가되는지 시험해보려 한다"며 "그러나 외국산 면역관문억제제는 가격이 비쌀 뿐만 아니라 이를 개발한 글로벌 제약사 입장에서 병용 투여 임상을 요청하는 업체들이 줄 서 있는데 굳이 한국 제약사와 공동 연구를 할 이유가 없다"고 말했다. 이에 국내 제약사들의 임상 수요를 충족하고, 값비싼 면역관문억제제를 건강보험에 등재해 국가 재정 부담을 덜기 위해서는 국산화가 반드시 필요하다는 설명이다. 그중에서도 `PD-1`과 `PD-L1`을 표적으로 하는 억제제 개발이 시급한 실정이다. 여러 가지 면역관문 가운데 암세포가 지닌 `PD-L1`이란 물질은 면역세포 브레이크의 일종인 `PD-1`이 발동하도록 해 면역세포를 약화시킨다. 또한 PD-L1은 암세포를 마치 면역세포인 양 둔갑시켜 면역체계를 교란한다. 박 대표는 "PD-L1을 보유한 암세포는 면역세포인 척 흉내내면서 면역세포 공격을 교묘히 피해간다"면서 "PD-1과 PD-L1은 암세포에서 보편적으로 발견되기 때문에 이를 표적으로 한 억제제를 개발하면 다양한 종류의 암을 정복할 수 있다"고 설명했다. 세계적으로 PD-1과 PD-L1에 달라붙어 기능을 차단하고 면역을 정상화하는 항체 의약품이 잇달아 개발되고 있는데, 최근 국내에서도 건강보험에 등재된 `옵디보`와 `키트루다`가 대표적이다. 박 대표는 "2022년이면 면역 항암제 시장이 30조원으로 성장할 것으로 예상되는데 옵디보와 키트루다가 전체 시장의 80%를 양분하고 있다"며 "두 제품의 건강보험 등재로 국가 재정 부담이 엄청나게 늘어날 텐데, 옵디보와 키트루다 특허가 모두 풀리는 게 2030년이라 그때까지 바이오시밀러(복제약)도 출시할 수 없다"고 말했다. 틈새시장을 겨냥하기 위해 와이바이오로직스의 면역관문억제제는 특허 만료 시점 이전인 2025년 출시를 목표로 개발이 진행 중이다. 박 대표는 "국내에서 PD-1과 PD-L1 항체를 보유한 회사가 거의 없다"며 "올해 이들을 표적으로 한 면역관문억제제 후보물질에 대한 전임상(동물실험)에 들어간 뒤 내년께 임상 1상을 시작하는 게 목표"라고 말했다. 이어 "공동 연구를 검토하는 제약사에서 우리의 항체가 기존 면역관문억제제 대비 동등 이상의 효과를 낸다는 연구 결과를 전달받은 만큼 출시되면 좋은 대안이 될 것"이라고 덧붙였다. 와이바이로직스는 자체적으로 구축한 항체 라이브러리와 고속 항체 검색 시스템을 통해 이미 PD-1과 PD-L1 항체를 발굴해냈다. 1000억개에 달하는 인간 항체 데이터베이스(DB) 플랫폼을 갖췄기에 가능했다는 설명이다. 박 대표는 "우리는 핵심 기반기술을 축으로 새로운 항체신약 파이프라인을 내놓고 용역 서비스를 제공하는 연구개발(R&D) 중심 회사"라며 "임직원 52명 가운데 44명이 연구개발 인력"이라고 설명했다. R&D 중에서도 특히 후보물질을 찾는 연구(Research)에 특화돼 있다. 대표적인 기반기술이 인간 항체 라이브러리다. 박 대표는 "마치 보물창고처럼 다양한 항체를 보유하고 있어 특정 질환을 치료하는 데 필요한 항체를 빠르게 검색할 수 있고, 항체를 아웃소싱하거나 외국에서 가져온 회사들과는 차별화된다"고 말했다. 항체마다 특정 항원을 인식하는 부위가 있는데 그 구조와 모양은 가지각색이다. 마치 열쇠처럼 손잡이 모양이 같을지라도 문을 여는 데 필요한 톱니는 조금씩 다 다른 것과 마찬가지다. 와이바이오로직스의 고속 항체 검색 시스템은 어떤 항체가 어떤 항원에 가장 잘 붙고, 질병 원인물질을 효율적으로 제거하는지 등을 파악해 최적의 후보물질을 찾아준다. 이중항체 기술인 `앨리스(ALiCE)`를 바탕으로 새로운 항체 치료제도 개발 중이다.최근 각광받고 있는 CAR-T 세포치료제의 단점을 극복한 신약이다. 면역세포인 `T세포`를 조작해 암세포만 공격하는 CAR-T 치료제는 높은 완치율로 주목받고 있지만 대량 생산이 어렵고 약물의 지속 시간이 떨어진다는 단점이 있다. 박 대표는 "앨리스 플랫폼을 활용하면 기존 항체보다 항원을 더 잘 인식할 수 있기 때문에 항원을 잡는 힘이 세지고, 약물이 혈액 속에 더 오래 머무를 수 있다"며 "하나의 항체로 암세포를 잡고, 다른 항체로 T세포를 활성화한다"고 말했다. 박 대표는 "이른 시일 내에 동물실험을 진행할 예정"이라고 밝혔다. 와이바이오, 이중항체 플랫폼 활용…
와이바이오로직스가 이중항체 플랫폼 기술을 활용한 새로운 항암제를 개발한다. 이 약은 대량생산이 어려운 CAR-T 세포치료제의 단점을 극복하고, '옵디보', '키트루다'같은 면역항암제 효과까지 모두 확보할 전망이다.박영우 와이바오로직스 대표는 30일 민영통신 '뉴스1'이 한국생명공학연구원 생명공학정책연구센터와 주최한 한국바이오포럼 2017에서 신규 플랫폼 기술 '앨리스(ALiCE)'와 신규 개발 항체치료제를 소개했다.이날 박 대표는 "최근 CAR-T 세포치료제가 높은 완치율로 시장의 각광을 받고 있는 상황이지만, 대량생산이 어렵다는 단점이 있다"라며 "우리가 개발한 플랫폼 기술 앨리스는 이런 점을 극복할 수 있다"고 밝혔다.와이바이오로직스의 앨리스는 기존 단백질 치료제 개발 기술인 '바이트(BiTE, Bispecific T-cell Engager)'를 개량한 이중항체 플랫폼이다. 기존 바이트는 생산 및 약물지속시간이 떨어진다는 단점이 있었으나 와이바이오로직스가 이를 개선했다.와이바이오로직스는 앨리스를 활용해 암 표적인자인 'PD-L1'을 목표로 하는 항체치료제를 개발할 계획이다. 앨리스를 사용하면 면역T세포와 결합하는 CD3 수용체와 암세포와 결합하는 이중 항체치료제를 만들 수 있다.박 대표는 "최근 세포실험을 통해 면역T세포를 활성화시키는 정도가 기존 항암제보다 월등히 높게 나타난 것을 확인했다"면서 "근시일 내로 동물실험에서 항암 항체치료제 'PD-L1 앨리스'의 동물실험을 진행할 예정"이라고 밝혔다.이 항체치료제가 상용화될 경우 PD-L1 표적항암제의 효과에 면역항암제가 가진 환자 본인의 자가면역 효과까지 더할 수 있을 전망이다. 특히 환자 개인 1명에 따른 치료가 아니라 대량 생산도 가능해 저렴한 가격까지 확보할 것으로 예상된다.박 대표는 "내년 5~6월께면 PD-L1과 앨리스를 결합한 신규 파이프라인을 선보일 수 있을 것"이라며 "시장에서 중거리 탄도미사일급 파급력을 가질 것으로 기대한다"고 말했다.한편, CAR-T 세포치료제는 현재 글로벌 의약품 시장에서 각광받는 항암신약이다. 환자의 말초혈액에서 면역세포인 T세포를 분리해 면역력을 강화한 뒤 다시 환자에게 주사하는 방식으로 1인 맞춤치료만 가능하다. LG출신 바이오 기업 CEO 3인방 뭉쳤다…동료 넘어
LG생명과학 출신 바이오 기업 최고경영자(CEO)들이 과거 한솥밥을 먹던 선·후배 사이를 뛰어넘어 최근 신약개발 사업파트너로 협력관계를 공고히 하고 있다. LG생명과학(현 LG화학)에서 선·후배 연구원으로 동고동락하다 각각 바이오 기업을 창업한 조중명 크리스탈 대표와 박영우 와이바이오로직스 대표, 김용주 레고켐바이오 대표가 의기투합해 신약개발 협력에 나선 것. 협력의 중심에는 항체의약품 개발 기술력을 갖춘 '와이바이오로직스'(대표 박영우)가 있다. 항체의약품(면역항암제 포함)은 최근 항암 신약개발 전선에서 가장 주목받고 있는 치료제 분야다. 표적항암제를 개발 중인 '크리스탈지노믹스'(대표 조중명)와 항체-약물 접합 기술(ADC, Antibody-Drug Conjugate)을 갖춘 '레고켐바이오'(대표 김용주)가 각각 와이바이로직스의 항체 의약품을 접목해 기존 약물들 보다 효과적인 항암제 개발에 나섰다.박영우 와이바이오로직스 대표는 "LG 출신이라는 공통분모가 서로의 신뢰를 쌓는데 영향을 미쳤고 무엇보다 각사의 기술경쟁력이 이번 신약 공동개발을 진행할 수 있게 했다"고 밝혔다. 이어 "전문 벤처들의 연합으로 난이도가 높은 신약개발에 막강한 힘을 발휘해 나갈 것"이라며 "다른 회사들에게도 이러한 협업모델이 본보기가 될 것"이라고 덧붙였다. 앞서 '크리스탈지노믹스'와 '와이바이오로직스'는 지난달 28일 각사의 대표 항암 신약물질로 병용요법 치료제를 공동개발하기로 합의했다. 면역항암제와 분자표적항암제를 병용투여할 경우, 표적치료제의 기폭제 효과로 면역항암제 효과가 더 좋아진다는 연구결과들이 최근 동물실험을 통해 밝혀지고 있어서다.'크리스탈지노믹스'는 암세포에 특이적으로 작용하는 자체개발 분자표적항암 신약물질 'CG200745'를 이번 병용치료법 개발에 활용할 계획이다. '와이바이오로직스'가 개발 중인 면역항암 항체는 체내 면역시스템을 활성화시켜 암세포를 공격하는 면역세포들을 활성화 시키는 차세대 항암요법제다. 구체적인 타깃 암질환은 임상단계를 거치면서 결정할 계획이다. '레고켐바이오'는 항체와 약물 접합기술(ADC)을 갖춘 바이오벤처로 항체는 다른 기업으로부터 도입하고 있다. 그중 '레고켐바이오'가 손잡은 한 곳이 '와이바이오로직스'다. ADC는 항체가 암세포를 찾아 들어가면 항체에 접합된 약물이 암세포만 공격할 수 있도록 하는 기술이다. 양사간 협력에 따라 '레고켐바이오'는 '와이바이오로직스'로부터 제공받은 항체를 자체 개발 약물에 접합시켜 현재 간암 치료제 개발을 위한 전임상(동물)을 준비 중이다.이들 3사가 서로 협력하게 된 데에는 CEO가 모두 LG 출신으로 바이오 기업을 창업한 공통분모가 크게 작용했다. 조중명 대표는 지난 1984년부터 LG생명과학(현 LG화학)의 전신인 럭키바이오텍 미국지사를 시작으로 대덕연구단지에 위치한 LG화학 바이오텍 연구소장을 역임하다가 2000년 크리스탈지노믹스를 창업했다. 1985년 박영우 대표가 LG화학 바이오텍에 입사한 뒤 럭키바이오텍 미국지사로 발령받으면서 두 사람은 인연을 맺었다. 박 대표는 미국지사에 근무하다가 박사과정을 밟은 뒤 1999년 국내연구소로 돌아와 2005년까지 항체 치료제 팀장과 그룹장을 맡았고 2007년 항체기업 와이바이오로직스를 설립했다. 김용주 레고켐바이오 대표는 조 대표의 후배이면서 박 대표의 선배다. 김용주 대표는 LG화학 신약연구소장을 맡아왔다. 줄곧 국내 연구소에 있었기 때문에 조 대표와 박 대표가 한국에 들어오면서 서로 만났다. 이때 연구소 총괄 헤드가 조 대표였다. 김용주 대표는 합성의약 전공을 살려 2006년 레고켐바이오를 설립했다. "이게 뭐처럼 보입니까?" 명함을 건네던 박영우 와이바이오로직스 대표가 작은 Y자 모양 그림을 가리켰다. 와이바이오로직스의 와이 아니냐고 했더니 `항체`란다. 항체는 바이러스나 세균 같은 항원을 물리치고 우리 몸속에 침입한 미생물에 대항해 세포 외부 자극을 유도하는 당 단백질을 말한다. 흔히 `B형간염 항체가 있다`거나 `예방접종 후 항체가 형성됐다`는 말을 하는데, 신약개발 과정에서는 종양 등 치료 타깃을 찾아가 결합하는 역할로 사용된다.미사일로 치면 센서 같은 역할이다. 박 대표는 "우리 몸의 면역반응과 시스템은 놀라울 정도로 잘 적응되어 있다"며 "특히 항체는 약의 성질로 볼 때 이보다 더 완벽할 수 없을 만큼 최정점에 있다. 항체를 대체할 후보가 없다는 뜻"이라고 설명했다. 와이바이오로직스는 작년 제약바이오 업계와 벤처캐피털(VC)들을 깜짝 놀라게 했다. 불과 6개월 만에 글로벌 블록버스터 면역항암제(PD-1 억제제)와 동등 이상의 효과를 내는 항체를 찾아내며 기술력을 입증한 것이다. 반응은 뜨거웠다. 작년에만 150억원의 투자를 유치했고, 제약바이오기업들의 러브콜이 이어졌다. 작년 말 범부처신약개발사업단 지원과제에 선정된 데 이어 지난달 28일에는 크리스탈지노믹스와 공동 연구개발을 추진하기로 했다는 뉴스를 발표했다. 기술력을 입증한 지 불과 1년, 이 회사와 제휴한 제약바이오기업은 CJ헬스케어, 녹십자 목암연구소, 안국약품, 레고켐바이오사이언스 등 6곳이나 된다. "2년의 시간을 줘도 딱 맞는 항체를 못 찾는 경우가 있거든요. 설령 찾았다고 해도 `최적화`하는 과정이 필요한데, 우리는 단번에 최적화된 항체를 찾아냈습니다. 1000억개가 넘는 `항체 라이브러리`를 구축했고, 사람이 하나하나 찾던 과정을 많은 부분 자동화했기 때문입니다." 이 회사는 우리나라에서 유일하게 인간 항체 스크리닝 자동화 장비와 기술을 보유하고 있다. 종전에 4~6개월씩 걸리던 항체 스크리닝을 2주일 만에 끝낼 수 있는 시스템이다. 박 대표는 "우리가 보유한 1000억개의 라이브러리는 글로벌 경쟁사 수십 곳과 견주어도 결코 뒤지지 않는다고 자부한다"며 "플랫폼 기술로 국내외 제약바이오업체와 협력하는 한편, 신규 면역관문 항체를 발굴하고 독자적으로 항체신약을 개발하는 데도 힘쓸 것"이라고 사업모델을 소개했다. 와이바이오로직스가 주목받는 또 다른 이유는 국내외 제약사들이 앞다퉈 면역항암제 연구에 뛰어들고 있기 때문이다. 제약산업 분석업체인 이밸류에이트파마는 면역항암제인 옵디보, 키트루다 등이 2022년 글로벌 의약품 매출 1위와 9위에 올라설 것으로 전망했다. 면역항암제는 암 환자의 면역력을 높여 암세포와 싸우게 하는 치료제다. 독성이 있어 정상 세포까지 파괴하는 1세대 화학항암제, 종양만 타깃으로 하지만 내성이 생기는 2세대 표적항암제와 차별화해 `3세대 항암제`로 불린다. 다양한 암에 적용할 수 있고 부작용이 적다는 장점이 있다. "우리가 공동으로 개발 중인 면역항암제, 자가면역질환 치료제를 비롯해 항체치료제 시장은 고령화가 심화될수록 커질 수밖에 없습니다. 매일 약을 먹는 대신 6개월에 한 번만 주사를 맞으면 된다고 생각해보세요. 일상생활을 하면서 항암치료를 받을 수 있고, 지미 카터 전 미국 대통령 사례처럼 고령자도 항암제 투여가 가능합니다. 두 가지 약을 병용 투여해 시너지효과를 높일 수도 있고, 새로운 물질을 붙여 약점을 보완할 수도 있죠. 여러 제약바이오기업들이 우리 회사에 관심을 보이는 이유입니다." 박 대표는 LG생명과학에서 20년, 생명공학연구원에서 10년 가까이 항체만 연구해온 전문가다. 2015년부터 와이바이오로직스 CEO를 맡아 차별화된 기술력을 확보했다. 그는 "지난 30년간 전임상 단계에서 최적의 항체를 발굴하는 데 올인한 `탐험가`였다면 이제 임상이라는 가보지 않은 길을 떠나게 됐다"면서 "임상을 잘 아는 전문가를 영입하는 등 `드림팀`을 꾸려 우리 항체 단독으로 끝까지 신약을 개발하는 것이 꿈"이라고 말했다. [Bio & Tech] A Treasure Trove of 100 Billion Antibodies for New Drug Development
“What does this look like?” asked Y-Biologics CEO Young Woo Park, as he handed over his business card, while pointing at a small Y-shaped image. When I guessed that it was the initial of Y-Biologics, he explained that it was actually an antibody. Antibodies are glycoproteins that trigger an immune response against antigens of viruses, bacteria, and other microbes invading our body. You have probably heard that some have “hepatitis B antibodies” or some people “developed antibodies after being vaccinated.” In the process of developing new drugs, antibodies are used to find and bind to certain targets such as tumors. They are comparable to the sensors in a missile, for example. “Our body’s immune system and response are surprisingly well adapted,” CEO Park explained. He noted that “antibodies are the pinnacle of drugs, as they can’t get any more perfect than that. That is, there are no other candidates that can substitute antibodies.” Y-Biologics left the biopharmaceutical industry and venture capitalists (VCs) last year by proving its technological prowess by finding an antibody that has an effect equal to or greater than that of a global blockbuster immuno-oncology agent (PD-1 inhibitor) within just six months. The news caused a huge sensation. The company received KRW 15 billion in investment last year alone and endless collaboration requests from pharmaceutical and bio companies. It was chosen by the Government-wide New Drug Development Project Group for research funding at the end of last year, and last month, the company announced that it would carry out a joint R&D project with Crystal Genomics. In just one year after proving its technological power, Y-Biologics has signed partnership agreements with six pharmaceutical and bio companies, including CJ Healthcare, Green Cross Institute for Biomedical Research, Ahn-gook Pharm, and Lego Chem Biosciences. “There are cases where the right antibody cannot be found even after two years. Even if it is found, there is a need to ‘optimize’ it. We, on the other hand, found an optimized antibody at once. This was possible because we had built an ‘antibody library’ of over 100 billion antibodies and automated many parts of the related processes that were performed manually,” said CEO Park. Y-Biologics is equipped with Korea’s one and only automated human antibody screening equipment and technology. The automated system has cut the antibody screening time from 4 to 6 months to just 2 weeks. CEO Park said, “I am confident that our library of 100 billion antibodies is comparable to the antibody libraries of dozens of global competitors,” and introduced the company’s business model, which is to “collaborate with domestic and overseas pharmaceutical and bio companies based on [its] platform technology and dedicate [itself] to discovering new immune checkpoint inhibitors and developing new antibody drugs independently.” Another reason Y-Biologics is garnering attention is the fact that domestic and foreign pharmaceutical companies are joining the race to develop immuno-oncology drugs. Evaluate Pharma, which analyzes the pharmaceutical industry, predicted that immuno-oncology drugs, Opdivo and Keytruda, would rank first and ninth in terms of global pharmaceutical sales in 2022. Immuno-oncology drugs are therapeutics aimed at enhancing the immune systems of cancer patients to better fight cancer cells. Chemotherapy agents are “first-generation anticancer agents” that are toxic and destroy even normal cells, while targeted therapy agents are “second-generation anticancer agents” that target only tumors but eventually lead to tolerance. Immuno-oncology drugs are called “third-generation anticancer agents” that are applicable to various cancers and cause few side effects. “The market for antibody therapeutics, including immuno-oncology and autoimmune disease drugs, which we are jointly developing with our partners, will grow inevitably, as population aging occurs at an accelerated rate. Imaging just having to receive an injection every six months, instead of taking medication every single day. It’s possible to receive treatment, without any disruptions in your day-to-day life, and as with the case of former U.S. President Jimmy Carter, the elderly can receive this type of anticancer treatment. By administering two drugs concomitantly, a synergistic effect can be achieved, and new substances can be added to address any drawbacks. This is why many bio and pharmaceutical companies are showing interest in our company.” CEO Park is an expert in antibody drugs who has been researching antibodies for 20 years at LG Life Sciences and for nearly 10 years at the Korea Research Institute of Bioscience and Biotechnology. Since 2015, he has been serving as the CEO of Y-Biologics. “I was an ‘explorer’ who went all-in to discover the optimal antibodies in the preclinical phase for the past 30 years, and now I am now on a path to the clinical phase, which I have never been on,” said CEO Park who then added that it is his “dream to develop a new drug until the end by recruiting experts on clinical research to form a ‘dream team.’” “The goal is to complete the Phase 1 clinical trial in 2019 for treatment of rare cancers, for which the Phase 3 clinical trial requirement is waived. If everything goes according to plan, we will be able to finish the clinical trial in 2023, or 2024, or perhaps even before then. We will also continue our research on ‘Better PD-1’ to improve the performance of our existing drug and bispecific antibodies that bind to two targets. CG200745는 서울아산병원에서 췌장암치료제 임상 1b/2상을 진행 중이다.면역관문억제제는 뛰어난 치료효과에도 불구학고 실제 환자에서의 반응율은 20%에 그친다고 알려져 있다. 그러나 T세포를 활성화해 면역력을 증진시키는 기전을 바탕으로, 기존 항암제와 병용투여할 경우 치료효과가 월등히 높아진 연구결과가 발표되면서 다양한 병용투여임상이 활발히 진행되고 있다. 박영우 와이바이오로직스 대표는 지난10일 (재)범부처신약개발사업단(KDDF)과 이중표적 자가 면역질환치료제 개발을 위한 `범부처전주기신약개발사업 과제` 협약을 체결했다고 11일 밝혔다.와이바이오로직스는 이번 과제선정으로 사업단의 지원을 받아 자가염증질환를 치료하는 이중항체개발에 속도를 낼 수 있게 됐다.이번 과제에 선정된 이중항체(YBL-004)는 염증을 유발하는 사이토카인(cytokine) TNF- α 와 IL-17의 활성을 동시에 저해하는 물질이다. Y-Biologics Receives KRW 5 B Worth of New Investment in Common Stock
Y-Biologics has laid the groundwork to speed up the development of new human antibody drugs. Young Woo Park, CEO of Y-Biologics, announced that the company has received an investment of KRW 5 billion in common stock. Intervest, which participated in the second round of funding made an additional investment of KRW 3 billion, while Mirae Asset Daewoo decided to additionally invest KRW 2 billion for a total of KRW 5 billion. It has been seven months since the company raised KRW 10 billion from seven institutions in the second round of funding in the beginning of this year. As a result, Y-Biologics has received a total of KRW 16 billion over three rounds of funding. Investors in the second round acquired redeemable convertible preferred stock (RCPS), while those in the third round received common stock. This past October, Y-Biologics garnered much attention for discovering PD-1 and PD-L1 human antibodies after just 6 months of research and licensing them to Ahn-gook Pharm and Green Cross MOGAM Institute for Biomedical Research. Based on its bispecific antibody platform technology, the human antibody R&D biotech company signed a contract with CJ Healthcare this past May to conduct joint global research on three bispecific antibodies. “We are actively conducting joint research in collaboration with bio startups, pharmaceutical companies, and hospitals in Korea,” said Bum Chan Park, Vice President of Y-Biologics, who then explained that “the scope of the research is not limited to antibodies, as we are capable of conducting joint research on CAR-T and CAR-NK, which are cell therapy agents requiring specificity.” Y-Biologics is actively conducting R&D to establish a “Better PD-1, PD-L1 franchise” with superior efficacy than the existing agents, based on bispecific antibodies that can recognize two antigens at the same time. The company raised KRW 10 billion in Series B funding in the first half of this year, and it is expected to receive an additional investment of up to KRW 20 billion within this year. The vast majority of those considering investing in the antibody drug developer in the second half of the year are venture capital companies that took part in the Series B funding in the first half. They are most likely to invest around KRW 10 billion, similar to the first half. So why are major domestic venture capital companies considering investing in Y-Biologics? The reviewers with expertise in investing in bio companies at venture capital companies say that it is due to Y-Biologics’ unique antibody drug candidate discovery technology that ensures a swift discovery process. There are also high expectations for mid- to long-term performance as the company is equipped with diverse and solid platforms, which is rare for bio startups. Y-Biologics, established in 2007, is an R&D company with most of its employees boasting extensive experience in participating in a number of large-scale national antibody drug development projects. It possesses core platform technologies, professional research personnel, and automation and analysis equipment that are essential for initial research for new antibody drug development, candidate antibody screening, and discovery of candidates for actual development. Ymax-ABL, the technology that sets the company apart from its peers, is the largest library in Korea that contains about 100 billion different antibodies. With Ymax-ABL as well as Korea’s first automated screening system and optimized biopanning technology, Y-Biologics is laying the groundwork for screening human antibodies essential for antibody drug development. Other differentiated technologies of Y-Biologics include Ymax-CEPT, a core technology for trawl-type target discovery, and Ymax-tEXPRESS, which accelerated the initial antibody sample production process by maximizing protein production in animal cells. Ymax-tEXPRESS, in particular, is over 20 times more efficient than the similar technologies developed by general pharmaceutical companies. Y-Biologics is endeavoring to discover candidates for the development of antibody drugs with excellent efficacy in treating intractable diseases, such as cancer and autoimmune diseases. It became the first company in Korea to discover candidates for the development of immuno-oncology antibodies (anti-PD-1, anti-PD-L1) and is pursuing technology transfer and joint development with Ahn-Gook Pharm and Green Cross MOGAM Institute for Biomedical Research among others. Y-Biologics has yet to begin marketing its products, so it is believed that once the necessary network is created, it will be able to gain a greater share of the domestic market and gain sufficient competitiveness in overseas markets. Intervest, Kolon Investment, Smilegate Investment, E & Investment, Magna Investment, and Timewise Investment, and others that invested a total of KRW 10 billion in the first half of this year seem to anticipate the growth of Y-Biologics amid the expansion of the antibody drug market. CJ Healthcare, which is working with Y-Biologics to develop a dual-target antibody to secure a high value-added new drug pipeline, has also become a strategic investor (SI) noting the bio startup’s development capability. Y-Biologics currently has seven new antibody drug and protein drug pipelines. Their goal is to secure at least two more clinical pipelines in 2018, starting with a bispecific antibody pipeline for the treatment of autoimmune diseases, which will enter the preclinical trial phase next year. A representative of Y-Biologics said, “We will pursue IPO on KOSDAQ in 2019 by securing additional pipelines and such and make a leap as a competitive company in the global market by establishing various development and sales networks. [...] Our mid- to long-term goals are to obtain domestic marketing approval for a couple of our new antibody drug products in 2025 and develop the approved products for the global market by 2030.” Meanwhile, venture capital companies looking to make an additional investment this year have noted the possibility of IPO and conclusion of licensing agreements with domestic and foreign pharmaceutical companies. This is because Y-Biologics’ new antibody drug development technology is an original technology enabling discovery of various antibodies and high profits can be expected if each of the dozens of antibodies discovered is licensed out or commercialized after clinical trials. An industry official said, “The specific investment size has not been confirmed yet, but it being reviewed at a level similar to that of the Series B funding. [...] Investing in [Y-Biologics] seems very appealing because of their prompt new drug candidate discovery process and licensing strategy.” Y-Biologics recorded KRW 680 million in revenue by providing research services last year. 항체신약 발굴 연구 전문기업 와이바이오로직스에 대한 벤처캐피탈 업계의 관심이 커지고 있다. 상반기 100억 원의 시리즈B 투자를 마무리한 데 이어 추가 투자 수요도 이어지고 있어 연내 최대 200억 원 규모의 투자가 가능할 전망이다. 하반기 투자를 검토중인 곳 중 대부분은 상반기 시리즈 B투자에 나섰던 벤처캐피탈이다. 펀딩 규모 역시 상반기와 비슷한 100억 원 안팎의 투자가 유력하다. 국내 주요 벤처캐피탈들이 와이바이오로직스에 대해 연이은 투자 검토를 추진하는 이유는 뭘까.벤처캐피탈의 바이오전문 심사역들은 와이바이오로직스만의 독특하고 신속한 항체 신약 개발 후보물질 발굴 기술을 꼽고 있다. 신생 바이오 벤처로는 드물게 다양하고 탄탄한 플랫폼까지 갖췄다는 점에서 중장기적 성과에 대한 기대감도 높다. 2007년 설립된 와이바이오로직스는 구성인력 대부분이 오랜 기간 다수의 대형 항체신약개발 국책과제에 참여해온 전문 연구 기업이다.특히 항체 신약 개발을 위한 초기 연구를 비롯해 후보항체 스크리닝 및 최종 개발후보 발굴에 필수적인 핵심 플랫폼 기술, 전문 연구인력, 자동화 및 분석 장비를 보유했다는 평가다. 와이바이오로직의 가장 차별적 기술인 Ymax-ABL 기술은 약 1000억 종의 서로 다른 항체가 포함된 국내 최대규모의 라이브러리 기술이다. Ymax-ABL은 국내 최초의 스크리닝 자동화 시스템과 최적화된 '바이오 팬닝' 기술을 통해 항체 의약품 개발에 필수적인 인간 항체를 선별하는 기반을 마련하고 있다. 저인망식 타깃발굴의 핵심기술 Ymax-CEPT과 동물세포에서 단백질 생산기술을 극대화해 초기 항체시료생산 프로세스를 가속화 시킨 Ymax-tEXPRESS 역시 와이바이오로직스의 기술 차별성을 높이고 있다. 특히 Ymax-tEXPRESS는 일반 제약사에 비해 20배 이상 높은 효율을 보이며 경쟁사 기술을 앞선다는 평가다. 난치성 질환인 암 및 자가 면역질환 등에서 탁월한 효능을 가진 항체의약품 개발후보를 발굴중인 와이바이오로직스는 국내 기업으로 처음 면역 항암제(항 PD-1, 항 PD-L1) 항체 개발 후보를 발굴하며 올해 안국약품, 녹십자 목암 연구소 등과 함께 기술이전, 공동개발 등을 추진중이다. 업계에서는 아직 와이바이오로직스가 본격적인 영업에 나서지 않은 상황인 만큼 향후 네트워크가 마련된다면 국내 시장 점유를 확대하고 해외 시장에서 충분한 경쟁력도 보유할 수 있을 것으로 평가하고 있다. 상반기 100억 원 규모의 투자에 나섰던 인터베스트와 코오롱인베스트먼트, 스마일게이트인베스트먼트, 이앤인베스트먼트, 마그나인베스트먼트, 타임와이즈인베스트먼트 등 역시 항체 신약 개발 시장의 확대 속에 이 같은 와이바이오로직스의 성장 가능성을 기대하는 모습이다. 고부가가치 신약 파이프라인 확보를 위해 이중타깃 항체 공동 개발에 나서고 있는 CJ헬스케어도 와이바이오로직스의 개발 능력에 주목해 전략적 투자자(SI)로 참여하고 있다. 와이바이오로직스는 현재 7개의 항체신약 및 단백질신약 파이프라인을 보유하고 있다. 내년부터 전임상에 진입하는 자가면역질환 치료용 이중항체 파이프라인을 필두로 2018년에는 임상 파이프라인을 2개 이상 확보하겠다는 목표다. 와이바이오로직스 관계자는 "파이프라인 확보 등을 통해 2019년 코스닥 시장에 상장하고 다양한 개발 및 영업 네트워크를 구축해 글로벌 시장에서 경쟁력있는 기업으로 도약할 것"이라며 "2025년에 국내에서 항체신약 제품 1~2개에 대한 허가를 이끌고 2030년에는 글로벌 시장내 허가 제품을 개발하는 것이 중장기 목표"라고 밝혔다. 한편, 연내 추가 투자를 검토하는 벤처캐피탈들은 와이바이오로직스의 상장과 국내외 제약사로의 기술이전 가능성에 주목했다. 와이바이오로직스의 항체신약 개발기술이 원천기술인만큼 다양한 항체 발굴이 가능하고 수십 개의 각 항체마다 별도의 기술 이전이나 임상 이후 사업화를 추진할 경우 높은 수익 창출이 기대되기 때문이다. 업계 관계자는 "아직 구체적인 투자 규모가 확정되지 않았지만, 시리즈 B투자와 비슷한 수준에서 투자를 검토하고 있다"며 "빠른 신약 물질 발굴과 기술이전 전략을 갖추고 있다는 점에서 투자 매력이 높은 상황"이라고 설명했다. 와이바이오로직스는 지난해 연구용역 대행을 통해 매출 6억 8000만 원을 기록했다. 최대주주는 박영우 대표로 지분 54.9%를 보유중이다. 와이바이오로직스, 녹십자와 면역항암제 공동 개발

와이바이오로직스·목암연구소, 면역항암제 공동개발
항체전문 바이오업체 와이바이오로직스가 목암연구소와 손을 잡고 면역항암제를 공동으로 개발한다.새 면역항암제는 일부 암종에서 치료 효능을 탐색하는 연구가 진행될 예정이다. 와이바이오로직스는 20일 자체 개발한 항체 YBL-006과 YBL-007을 목암연구소에 기술이전하는 계약을 체결했다. 새 기술을 이전받은 목암연구소는 앞으로 자사가 보유한 항암치료 물질에 두 항체를 접목하는 방향으로 전임상(독성검사)을 진행할 계획이다.와이바이오로직스 역시 두 항체를 가지고 별도의 면역항암제 개발을 진행할 예정이며 현재 전임상 단계를 설계 중이다.회사 측은 지난달 안국약품과 면역항암제 개발에 관한 계약을 체결했고 기술을 이전할 또 다른 업체를 물색 중이다. 최승현 목암연구소장은 "면역항암제는 차세대 항암제로 주목받고 있다"며 "신약 개발에 속도를 내겠다"고 밝혔다.박영우 와이바이오로직스 사장은 "유망 항체를 발굴하는 데는 국내에서 독보적인 경쟁력을 확보했다"며 "다양한 면역항암제 개발이 이뤄지도록 하겠다"고 말했다. "이미 확보한 인간항체 라이브러리만 1000억 여종이 넘습니다. 앞으로 국내에서 인간항체를 개발하기 위해서는 우리 회사와의 연계가 필수적일 것입니다."박영우 와이바이오로직스 대표는 21일 서울 영등포구 여의도동 미래에셋대우 본사에서 열린 바이오투자포럼에서 회사가 가진 핵심 경쟁력을 인간항체 라이브러리라고 소개했다.항체 신약은 항체가 특정 물질에 특이적으로 결합하는 성질을 이용한 것으로 기존 치료제가 정상세포를 공격하면서 생기는 부작용을 줄이며 동시에 치료효능을 높인 2세대 단백질의약품이다.박 대표는 "1997년 최초의 항체 의약품인 레미케이드가 출시되면서 현재 글로벌 탑 10에 속하는 의약품 중 7개가 항체치료제"라면서 "특히 인간항체가 가장 주목받는데 우리는 골수에서 유래한 면역세포로부터 인간항체 라이브러리를 확보하는 독보적인 기술을 갖고 있다"고 소개했다.박 대표는 LG생명과학에서 항체 치료제 팀장으로 14년간 단백질 의약품을 개발한 경험을 갖고 있다. 이후 한국생명공학 연구원에서 다수의 항체신약개발 국책과제를 수행했다. 이러한 경험을 바탕으로 그는 2000년 초부터 ‘인간항체 라이브러리 구축’이라는 목표를 갖고 현재 7개의 파이프라인을 진행하고 있다.박 대표는 “국내에서 유일하게 하루에 만개 이상의 인간항체 스크리닝이 가능한 자동화 시스템을 갖추고 있다”라며 “우리 기술이 글로벌 수준에서도 대등하다는 것을 최근 각광받고 있는 면역관문억제제 PD-1, PDL-1 항체를 만들어 입증했다"고 강조했다.와이바이오로직스에서 가장 빠르게 진행되고 있는 파이프라인 YBL-006, YBL007은 각각 PD-1, PDL-1을 겨냥하는 항체로 올 초부터 시작해서 4개월 안에 항체 제작을 완성했으며 기존 항체와 동등한 성능을 나타내는 것을 확인했다.와이바이오로직스는 이러한 경험을 바탕으로 앞으로 ‘Better PD-1 혹은 PDL-1’을 개발할 계획이다. 특히 PD-1, PDL-1을 가지면서 동시에 면역활성화 물질인 사이토카인을 조절하는 등의 새로운 기능을 포함한 이중항체 YBL-004와 YBL-005는 곧 비임상을 들어갈 예정이다.그는 “YBL-004는 국가연구과제로 임상 1상까지 지원받을 예정”이라며 “Better PD-1, PDL-1을 위한 후보물질 탐색은 계속적으로 확대해 나갈 것”이라고 밝혔다.YBL-001~003은 완전혁신 신약으로 추가적인 기전연구를 진행중이다. Y-Biologics Builds Human Antibody Library
“We have already built a human antibody library of more than 100 billion types of antibodies. In the future, a partnership with our company will be essential for domestic companies to develop human antibodies.” Young Woo Park, CEO of Y-Biologics, introduced the human antibody library as the core competitiveness of the company at a bio investment forum held at the Mirae Asset Daewoo headquarters in Yeouido-dong, Yeongdeungpo-gu, Seoul. Novel antibody drugs are being developed immunotherapy based on the property of antibodies to specifically bind to a specific substance. Called second-generation protein drugs, antibody drugs are designed to exhibit high therapeutic efficacy, with the side effect of attacking normal cells – a problem exhibited by existing anticancer agents – minimized. “With the release of Remicade, the first antibody drug, in in 1997, seven of the ten top-selling drugs in the world are currently antibody therapeutics. [...] Human antibodies, in particular, are garnering the most attention, and we have a proprietary technology to secure a library of human antibodies from immune cells derived from the bone marrow,” said CEO Park. The CEO of Y-Biologics gained 14 years of experience in developing protein drugs as the head of an antibody therapeutics team at LG Life Sciences. Afterwards, he undertook multiple national projects for the development of novel antibody drugs at the Korea Research Institute of Bioscience and Biotechnology. Based on these experiences, he has been working on seven drug pipelines with the aim of building a human antibody library since the early 2000s. “Y-Biologics has Korea’s one and only automated system capable of screening more than 10,000 human antibodies per day,” said CEO Park before emphasizing that the company has “proven its technology is comparable to those of global companies with the development of PD-1 and PDL-1 antibodies, which are immune checkpoint inhibitors that are currently in the spotlight.” The pipelines that are being developed most quickly at Y-Biologics are YBL-006 and YBL007, which are antibodies targeting PD-1 and PDL-1, respectively. The development projects kicked off at the beginning of 2021, and the antibodies were produced in just four months. It has been confirmed that their performance is equivalent to that of existing antibody drugs. Based on this experience, Y-Biologics plans to develop “Better PD-1 or PDL-1.” Non-clinical research is set to begin in the offing with respect to the bispecific antibodies, YBL-004 and YBL-005, that will have additional functions such as regulating cytokines, which are immune activating substances, aside from binding to PD-1 and PDL-1. CEO Park revealed that “support will be received for the Phase 1 clinical trial of YBL-004 as a national research project” and that there are plans to “continue searching for candidate substances for Better PD-1, PDL-1.” YBL-001 to 003 are first-in-class drugs, for which additional research is underway to examine the mechanisms of action. YBL-001 is an antibody-drug conjugate (ADC) that Y-Biologics is conducting a joint study on with LegoChem Biosciences. The drug indications include brain cancer, liver cancer, and recurrent and metastatic cancer. YBL-003, on the other hand, is a novel immuno-oncology antibody drug targeting an immune checkpoint inhibitor. A competitive advantage that Y-Biologics has stems from its active collaboration with other pharmaceutical companies. To develop therapeutic agents its proprietary antibody library, joint R&D is being pursued with a number of pharmaceutical companies, including CJ Healthcare, LegoChem Biosciences, and Sanofi.
