와이바이오, 국립암센터와 치료용 항체 개발 MOU
항체기반 기술 지원 및 임상협력 와이바이오로직스(Y Biologics)는 10일 국립암센터 연구소와 치료용 항체 개발 상호협력을 위한 업무협약(MOU)을 체결했다고 밝혔다.이번 협약을 통해 와이바이오로직스는 국립암센터 연구소에서 추진중인 고형암 면역항암제 등의 연구에 항체 기반 치료기술을 지원하기로 했다. 국립암센터 연구소는 와이바이오로직스에서 개발한 항체의약품 임상개발에 협력할 예정이다.박영우 와이바이오로직스 대표는 “항암신약 개발 경험과 역량을 축적해온 국립암센터 연구소와 이번 협약을 계기로 암 정복을 향한 의지를 공고히 하고 더 큰 도전을 함께하길 기대한다”고 말했다.김영우 국립암센터 연구소장은 “항체 공학 기술과 라이브러리를 가지고 있는 와이바이오로직스와의 업무협약으로 항암신약 개발을 위해 협력해 나가겠다”고 밝혔다.와이바이오로직스는 항체 플랫폼 기술 기반의 신약개발 바이오벤처다. Y-Biologics Begins Dose Expansion Study for PD-1 Antibody in Australia
Set to evaluate the drug safety, efficacy, optimal dose, etc. with 70 solid cancer patients Y-Biologics, a biotech company specializing in the development of antibody drugs, has started administering YBL-006, a PD-1 immunotherapy candidate, in the Phase 1 dose expansion cohort in Australia. As was the case with the dose escalation study, the expansion cohort study will also first begin at Macquarie University Hospital. Y-Biologics received approval for Phase 1 clinical trials on YBL-006 in Korea and Australia in May last year and has since conducted a dose escalation study. The company announced the results of the phase 1, first-in-human clinical trial of solid cancer at the American Society of Clinical Oncology (ASCO) held this past June. The interim results confirmed drug safety and tolerability and demonstrated the clinical benefits of the drug, with one patient showing a complete response (CR). The dose expansion study, which recently commenced in Australia, is being conducted as part of a global clinical trial being carried out in Korea and Thailand among others. The objective is to evaluate the safety and efficacy of YBL-006 and the recommended Phase 2 dose (R2PD) of the drug for various types of solid cancer. This dose expansion study will proceed with around 70 solid cancer patients. Y-Biologics is aiming to market the first domestically licensed PD-1 antibody targeting rare types of cancer Korea and is developing PD-1 antibody in consideration of pursuing combination therapy with an immuno-oncology drug. Meanwhile, Y-Biologics is getting ready for its IPO in the second half of this year. The company selected Korea Investment & Securities as the underwriter and applied for a preliminary review in May. 와이바이오, "PD-1 항체" 호주 1상 "용량확장 시작"

Y-Biologics L/O Antibody for Immunotherapy to Pierre Fabre
The first accomplishment following a three-year joint research and license option agreement signed last year Reactivating immune cells in YBL-003 tumor microenvironment for cancer cell death △Young Woo Park, CEO of Y-BiologicsY-Biologics, a biotech company that develops antibody drugs, has signed a license agreement with French pharmaceutical group, Pierre Fabre, for exclusive development and commercialization rights to YBL-003, a new antibody candidate targeting solid cancer. According to the agreement, Y-Biologics will receive up to KRW 116.4 billion, including a non-refundable upfront payment, milestone fees for preclinical and clinical development stages, commercialization, and sales. Royalties will be additionally received according to the sales volume. YBL-003 licensed out by Y-Biologics is an IgG-based immune checkpoint inhibitor. However, the exact target was undisclosed. According to Y-Biologics, YBL-003 has a mechanism to regulate the function of macrophages and T cell activity to reactivate immune cells in the tumor microenvironment (TME) and kill cancer cells. It is expected that the scope of the indications will be expanded to include solid tumors such as stomach cancer, lung cancer, and breast cancer. The recent agreement is the first fruit of the joint research between the two companies that kicked off last year. The term of the joint research contract is 3 years, but it may be extended for 2 additional years. According to the terms and conditions of the contract, three targets will be examined each year, meaning that fifteen targets may be jointly researched and licensed during the maximum contract period of 5 years. Y-Biologics discovered a new antibody from Ymax®-ABL, a human antibody library, and Pierre Fabre verified that the antibody has the functional properties of an immune checkpoint inhibitor that reactivates a major immunosuppressive mechanism in the TME. 와이바이오, 피에르파브르에 "면역항암 항체" 1160억 L/O

와이바이오로직스, 과기정통부 지정 ‘2021 상반기 우수 기업연구소’로 선정

Y-Biologics is a startup that develops new antibody drugs using a human antibody library and bispecific T cell engager technology. Since May last year, it has been developing Korea’s first PD-1 antibody for immunotherapy in a clinical setting. The interim results of the current multinational clinical trials will be presented at the conferences of the American Society of Clinical Oncology and European Society for Medical Oncology this year. Since May last year, Y-Biologics has been conducting multinational Phase 1 clinical trials to examine the safety and efficacy of YBL-006, a PD-1 antibody, in solid cancer patients. The PD-1 antibody is an immuno-oncology drug that binds to the PD-1 protein, which is the immune checkpoint of T cells, and causes cancer cells to die by preventing them from evading the attacks from the immune system. Cancer cells express a protein called PD-L1 on their surface and bind to the PD-1 protein on the T cells that are trying to attack them. When an antibody is attached to PD-1 to prevent this, the T cells can attack and kill cancer cells as normal. MSD’s pembrolizumab (product name: Keytruda), which is dominating the PD-1 antibody market, has been approved by the U.S. FDA for indications for 26 types of cancer and has already become a primary prescription drug for anticancer treatment, with sales reaching USD 14.4 billion last year. Experts predict that it will become a standard anticancer drug in the future, as approval is being granted for combination therapy with various anticancer drugs one after another. Noticing the success of pembrolizumab, a number of multinational pharmaceutical companies are making an effort to secure PD-1 antibodies by directly developing them or licensing them from developers. This is evidenced by an increase in technology transfers of PD-1 antibodies. Development Background One of the core capabilities of Y-Biologics is its ability to discover target antibodies from its own human antibody library, Ymax-ABL, with high efficiency. A PD-1 antibody was selected as a candidate for a new antibody drug by using the library, and it began to be developed in full swing in 2018. A PD-1 antibody is a basic drug for immunotherapy, and it is one of the drugs that are most actively researched to check the possibility of being administered concomitantly with various types of anticancer drugs. This is because it is judged that it is difficult to penetrate the market that is already dominated by multinational pharmaceutical companies, such as MSD and BMS, with a PD-1 antibody without any differentiated characteristics. If Y-Biologics develops a drug with efficacy equivalent to or higher than that of Keytruda, currently the No. 1 in the market, by applying a new biomarker or combination therapy to the types of cancer with unmet needs, it may become the first domestically developed PD-1 antibody immuno-oncology drug to be marketed, and this will be an important milestone in the development of new anticancer drugs in Korea. As for the Phase 1 clinical trial of YBL-006, there are plans to confirm the types of cancer to examine in the Phase 2 clinical trial in the future dose expansion study and collect more clinical data. Antibody Properties When Y-Biologics screened for an PD-1 antibody from the antibody library, YBL-006 was derived based on the binding affinity to the antigen, PD-1, and the productivity. The level of the binding affinity of the PD-1 antibody to the PD-1 protein on the surface of T cells is directly associated with the immune checkpoint inhibitory effect of the antibody and T cell activation. This has a significant impact on the clinical efficacy from the aspect of pharmacodynamics. YBL-006, which was selected from among antibody candidates with strong binding affinity to PD-1, differs substantially from the representative PD-1 antibodies that are currently on the market such as pembrolizumab and nivolumab (product name Opdivo) in terms of antigen-antibody binding properties, and the dissociation rate is significantly low, based on which outstanding efficacy can be expected. The reason behind these binding properties of YBL-006 can be seen by examining the region where it binds to PD-1 by X-ray spectroscopy. As shown in the figure on the right, the PD-1 antibodies that bind to the PD-1 protein in blue have different binding sites (epitopes or antigenic determinants).YB-006 binds to the area that is most similar to the area where PD-L1 on cancer cells is attached, and the area of the binding site was measured to be 2, 334 A², which is wider than pembrolizumab’s 2,126 A². This signifies that it has an excellent ability to expose cancer cells to the immune system attacks by inhibiting the immune checkpoints that work by binding to PD-1/PD-L1. Interim Results from Clinical Trials Y-Biologics has been conducting Phase 1 clinical trials on YBL-006 in Korea, Australia, and Thailand since May last year after completing a non-clinical trial in Canada. A dose escalation test has been completed to determine an appropriate dose, and it has recently begun a dose expansion study to create more clinical cases for specific types of cancer. In the dose escalation study, eleven patients with solid cancer, such as squamous cell cancer and colorectal cancer, participated, and the dose was increased to 0.5, 2, 5, or 10 mg/kg depending on the weight of the subject. As for the adverse events associated with the drug, one case of Grade 2 cytokine syndrome was reported, aside from fatigue and hypothyroidism, and no dose-based toxicity was observed. As such, it was deemed that there were no safety issues. The preliminary efficacy to be reported from the clinical trial was favorable. One subject showed complete remission (CR), with the disappearance of the target lesion, i.e., the tumor. Another subject showed partial remission (PR), with the tumor size reduced to 50%, providing evidence of the clinical efficacy of YBL-006. Two other subjects were classified under stable disease (SD), while the rest of the subjects were judged to have progressive disease (PD). Pharmacokinetic evaluation was also performed in the clinical trial. Based on the results, it was deemed reasonable to apply a single dose such as 200 mg and 300 mg, instead of the initial dose from Phase 1 clinical trial, which was 2 mg/kg at Week 2 or Week 3, like the existing PD-1 antibody drugs. There are plans to confirm the types of cancer to be targeted using YBL-006 in the Phase 2 clinical trial by conducting a dose expansion test and to collect more clinical cases. Future Clinical and Licensing Plans With the goal of developing Korea’s first PD-1 antibody for immunotherapy, Y-Biologics will initiate a Phase 2 clinical trial targeting rare cancers with a small number of patients first. Then, a protocol for applying new biomarkers is also being considered. This is a strategy to have YBL-006 be designated for the fast track process by the Ministry of Food and Drug Safety to facilitate the conditional approval. Under circumstances, obtaining this approval from the Ministry of Food and Drug Safety with multiple PD-1 antibodies already developed will facilitate licensing out in the conditional permit visibility after being designated as a target of expedited review by the Ministry of Food and Drug Safety. In a situation where a considerable number of PD-1 antibodies have already been developed, it is to more abundantly meet the requirements for license-out through the approval of the Ministry of Food and Drug Safety. PD-1 항체 시장을 선도하는 MSD의 ‘펨브롤리주맙(제품명 키트루다)’은 이미 26개의 암종에 대한 적응증을 FDA로부터 허가받았고, 작년 매출이 144억 달러에 이를 만큼 이미 항암치료의 주처방 약제가 되어가고 있다. 전문가들은 다양한 항암제들과의 병용요법도 속속 허가를 받고 있어 향후 표준요법 항암제가 될 것으로 전망하고 있다.이러한 펨브롤리주맙의 급성장에 자극받은 여러 다국적 제약사들은 자체적으로 개발하거나 도입하여 PD-1 항체를 확보하려는 노력을 기울이고 있으며, 최근 이를 증명하듯이 PD-1 항체들의 기술거래가 매우 활발하게 일어나고 있다.개발 배경와이바이오로직스는 자체 보유한 인간항체 라이브러리 ‘Ymax-ABL’로부터 타깃 항체를 고효율로 발굴해내는 것을 핵심역량으로 갖고 있다. 이를 적용한 항체신약 후보로 PD-1 항체를 선정하고 2018년부터 본격 개발에 착수했다. PD-1과 결합력이 강한 후보 항체들 중에서 선별한 YBL-006은 항원-항체 결합 특성이 펨브롤리주맙이나 니볼루맙(제품명 옵디보)과 같은 기존의 대표적인 PD-1 항체들과 많이 다르고, 특히 결합 후 해리되는 속도가 현저히 낮아 우수한 효능을 나타낼 것으로 기대된다.이와 같은 YBL-006의 결합 특성은 X-선 분광학으로 PD-1 단백질과의 결합 부위를 조사해보면 그 이유를 파악해 볼 수 있다. 오른쪽 그림과 같이, 파란색의 PD-1 단백질에 결합하는 PD-1 항체들은 그 결합부위(항원결정기, epitope)가 서로 다른데, 그중 YBL-006은 암세포의 PD-L1이 붙는 부위와 가장 유사한 부위에 결합하면서 그 결합 부위의 면적도 펨브롤리주맙의 2126A²보다 넓은 2334A²으로 측정됐다. 이는 PD-1/PD-L1 결합으로 작동하는 면역관문을 억제해서 암세포를 면역계의 공격에 노출시키는 능력이 우수하다는 것을 의미한다.임상 연구 중간 결과와이바이오로직스는 캐나다에서 비임상 시험을 마친 후 작년 5월부터 한국, 호주 그리고 태국에서 YBL-006의 글로벌 임상 1상 시험을 진행하고 있다. 현재 적정한 투여량을 확인하는 용량상승시험을 마치고 특정 암종에 대해 보다 많은 임상 증례를 만드는 용량확장시험에 진입하고 있다.용량상승시험에는 편평세포암이나 대장암 등 11명의 고형암 환자가 피험자로 참여했고 피험자의 체중에 따라 0.5, 2, 5, 10mg/kg으로 용량을 늘려가며 시험을 마쳤다. 약물과 관련된 부작용은 피로나 갑상선 기능 저하증 외에 2등급의 사이토카인 증후군 1건 정도가 집계됐고, 용량에 따른 독성도 발견되지 않아 안전성에는 문제가 없는 것으로 확인됐다.또 이 임상시험에서 보고자 한 예비유효성도 긍정적이다. 1명의 피험자에게서 완전관해(CR)가 나타나 표적병변, 즉 종양이 사라졌다. 다른 1명의 피험자는 종양이 50% 수준으로 줄어든 부분관해(PR)를 보여 YBL-006의 임상적 효능을 확신할 수 있었다. 그 밖에 2명의 피험자는 안정병변(SD)을 보였고, 나머지 피험자들은 병기진행(PD)으로 판정됐다. 이 시험에서는 약동학 평가도 함께 진행되었다. 기존의 PD-1 항체 약제들과 같이 2주 또는 3주의 투약 주기에, 2mg/kg과 같은 초기 임상 1상 시험 용량이 아닌 200mg이나 300mg과 같은 단일 투여량을 적용하기에 무리가 없는 것으로 판단됐다.YBL-006의 임상 1상 시험은 향후 용량확장시험에서 임상 2상 시험에 적용할 암종을 확정하고 보다 많은 임상 증례를 수집할 계획이다. 향후 임상 및 허가 계획와이바이오로직스는 국내 최초의 PD-1 항체 면역항암제의 개발을 목표로 우선 환자수가 적은 희귀 암을 대상으로 임상 2상 시험을 수행할 계획이다. 또 신규 바이오마커를 적용하는 프로토콜도 함께 고려하고 있다. 식품의약품안전처의 신속심사대상으로 지정 받아 조건부 허가 가시권에 YBL-006을 넣기 위한 전략이다. 적지 않은 수의 PD-1 항체가 이미 개발된 상황에서 식약처 허가를 통해 라이선스아웃의 요건을 보다 풍부히 갖추기 위해서다. 최지원 기자

Venturing into Immuno-Oncology Market with Antibody Library and Bispecific Antibody Platform
Heo-terview | Young Woo Park, CEO of Y-Biologics “We plan to proceed with an expansion study on YBL-006, an immuno-oncology drug that has entered a Phase 1 clinical trial” In response to the genetic engineering boom in the 1980s, LG Life Sciences (present-day LG Chem) established a genetic engineering research institute in Oakland. Park who was dispatched there returned to Korea in 1999 after mastering various genetic engineering techniques for 14 years and endeavored to develop new drug called antibodies. Since Janssen received marketing approval from the U.S. Food and Drug Administration (FDA) for Orthoclone, a therapeutic agent for kidney transplant rejection, in 1985, he was among the first in Korea to begin antibody drug research. After LG Life Sciences shut down its antibody drug development operations, Park joined the Korea Research Institute of Bioscience and Biotechnology where he studied antibodies for about a decade before establishing a startup to develop immuno-oncology drugs. We met with Young Woo Park, the founder and CEO of Y-Biologics, who is preparing to take another leap forward for new drug development through an IPO by filing a preliminary review application to the Korea Exchange, to hear the story of Y-Biologics, a developer of antibody drugs. Q. You were among the first in Korea to study antibody drugs. I am curious as to why you got into antibody research. A. I got my first real job in 1985 at the LG Chem Research Center located in Jeonmin-dong, Daejeon. Back then, there was a boom of genetic engineering, and large corporations were establishing genetic engineering research institutes. So did LG Chem in Daejeon. In Korea, at the time, genetic engineering technology was not up to the standard, so LG established a local research institute in Oakland near San Francisco. I returned to Korea around 1999 when LG commenced an antibody project. From 1997 to 2000, blockbuster antibody drugs were being developed, and LG also jumped in. Considering that drug development in Korea centered on small molecules at the time, it was a bold move. I got to learn many new things while doing research on antibodies at LG and the Korea Research Institute of Bioscience and Biotechnology (KRIBB). What was certain was that it was worth giving a shot at building a library of antibodies, compared to small molecules, as long as one had excellent technology because the scale did not matter. So I came to the conclusion that even a startup could give a shot at this. Q. That was a great segue, explaining why established a startup. Domestic startups focusing on development of single pipeline and new drug development models using antibody libraries sound somewhat new. A. I have been conducting research using phage technology since the time I was doing my master’s. Based on that experience, I conducted molecular cloning research at LG. With the research experience I gained at LG and KRIBB, I was confident when it came to synthesizing antibodies. Of course, in order to explain this to someone, a process of providing proof was necessary. So, to demonstrate, we went through the process of checking the diversity of the antibodies we had using next-generation sequencing (NGS) three to four years ago. In other words, our antibody library exists in the form of DNA, and we checked the sequence diversity through an antibody phagemid sequencing process. The higher the antibody diversity, the better the library quality. In this process, we were able to discover an excellent PD-1 antibody, YBL-006, for which a Phase 1 clinical trial is currently underway. Y-Biologics’ antibody library, Ymax-ABL [Source: Y-Biologics website]Q. Please explain YBL-006 in detail. A. The global PD-1 antibody market is currently worth KRW 30 trillion. The market is dominated by Keytruda (pembrolizumab) and Opdivo (nivolumab). We are conducting clinical research to get into this market. We have carried out a Phase 1 trial with 11 subjects, and we plan to increase the number of subjects to 50 through an expansion study. In the case of the clinical trial with 11 subjects, it was a process of checking the safety in terminal cancer patients, irrespective of the type of cancer they had. Once the results from the expansion study with 50 subjects come out, we will present them at the European Society for Medical Oncology (ESMO). Q. The PD-1 immuno-oncology drug (antibody) market is already dominated by Keytruda and Opdivo. What sets YBL-006 apart from others? A. When the physical properties were tested during the preclinical process, it was found that the properties of YBL-006 were much superior. YBL-006 exhibited five times better binding affinity than Keytruda. The area where PD-1 and PD-L1 ligands are bound by YBL-006 is much larger compared to Keytruda, based on which we can expect high efficacy. Q. You said that the Phase 1 clinical trial was conducted with 11 subjects with all kinds of cancer. What type of cancer will be targeted in the expansion study? A. In order to speed up the development process, relatively speaking, we will target rare types of cancer. We have formed a scientific advisory board (SAB) with the CEO of Bang & Ok Consulting, Professor Eui Cheol Shin from KAIST Graduate School of Medical Science and Engineering, Professor Woo Ik Jang who has worked at CHA Hospital and Handok, and Professor Seung Hoon Lee who was at the National Cancer Center and is now at Daejeon Eulji Medical Center. We are discussing the SAB about which the types of cancer we will target in our subsequent clinical trials. Since there are already similar drugs out on the market, the goal is to obtain approval as quickly as we can by demonstrating clear effectiveness. We signed an MOU with Samsung Medical Center in Seoul to pave the foundation for follow-up clinical trials. We are also discussing the matter with the National Cancer Center. Q. You have partners for other pipelines but not for YBL-006. Do you have a specific timepoint to license out the technology in mind? A. After the Phase 2 clinical trial, we will begin considering licensing out the technology. Also, research on combination therapy with the PD-1 antibody and other anticancer drugs is being carried out in full swing, so this will be our research strategy. Q. It seems that such active collaborations have been possible since you have such an excellent antibody library. Could you explain your collaboration strategy? A. CAR-T therapeutics, antibody-drug conjugates (ADCs), and T cell engagers are similar in that they are designed to control cancer cells. Ultimately, the key is to accurately target cancer cells. Antibodies play a crucial role in the target process. Once a suitable antibody is discovered, it ultimately becomes possible to effectively develop a CAR-T cell therapy agent, ADC, and T cell engager. Although there have been active discussions about T cell engagers, they have yet to be used widely as a drug is because there are hurdles caused by the toxicity issue and so on. This can be addressed with antibodies. This is the reason we are conducting joint research with LegoChem Bioscience, Intocell, and AbTis, which have ADC-related technology. Our company is equipped with a bispecific antibody platform called ALiCE and an antibody library called Ymax-ABL. In addition, we will file a patent registration application after developing a platform that can target and kill only tumor cells. △Y-Biologics’ bispecific antibody platform, ALiCE [Source: Y-Biologics website Q. So there can be a platform that can target and kill only tumor cells? I’d like to know more. A. This is a strategy to leverage the activity of the antibody by using the tumor microenvironment (TME). Startups abroad have jumped into the platform development race, and big pharmaceutical companies are buying these technologies. Research for a more precise effector function in the TME is being carried out in full swing as well. To be more specific, antibody activity can vary depending on the pH and proteases, which are enzymes that break down proteins, and the strategy is to pick out antibodies that can function in the relevant environment. The ultimate objective is to target only cancer cells without touching normal cells. These kinds of antibodies will be useful to companies that develop ADC and CAR-T therapeutics. It was possible for us to develop this type of platform because we had the bispecific antibody platform, ALiCE, and the antibody library, Ymax-ABL." Q. It may be too early to tell, but do you have a specific plan for raising capital after going public? A. Based on solid preclinical research, we will pursue in-house development until at least Phase 1. Then, we will seek to license out the technologies afterwards. When it comes to antibodies, cell line development and sample production are very costly. We will reinvest the funds to further expand our pipelines. Q. What are your ultimate aspirations as a startup specializing in new drug development? A. I’ve heard that Keytruda or Opdivo are not accessible to all patients due to the limited health insurance coverage in Korea. These drugs are used in primary treatment for non-small cell lung cancer which is quite prevalent, so I understand the sufferings of the patients and the concerns of the authorities. "1985년 첫 사회생활을 시작한 곳이 대전 전민동에 위치한 LG화학 연구소였어요. 당시 유전공학 붐이 일면서, 대기업에서도 적극적으로 유전공학 연구소를 설립했거든요. LG화학도 당시 대전에 연구소를 설립했는데, 아직 국내는 이렇다할 유전공학 기술이 없어 LG는 미국 샌프란시스코 근교 오클랜드에 현지 연구소를 설립했어요. 14년 간 현지 연구소에 파견돼 연구 생활을 이어갔어요.LG가 항체 프로젝트를 시작할 1999년 무렵 한국으로 귀국했어요. 1997년에서 2000년대는 블록버스터 항체 의약품이 개발되던 시기였는데, LG도 여기에 뛰어든 것이죠. 저분자화합물(small molecule) 중심으로 이뤄지던 당시 국내 신약개발 환경을 생각하면, 대단한 도전이었습니다.LG와 한국생명공학연구원(생공연)에서 항체 연구를 하며 많은 것을 접할 수 있었습니다. 확실한 것은 저분자화합물 대비 항체 의약품 라이브러리는 규모가 아니라, 우수한 기술력으로 도전해 볼 만하다는 것이었죠. 벤처도 충분히 도전해 볼 수 있는 영역이라는 판단이 섰습니다."Q. 자연스럽게 창업 이유까지 이유까지 설명해 주셨네요. 단일 파이프라인 중심으로 개발에 임하는 국내 벤처들과 항체 라이브러리를 활용한 신약개발 모델은 좀 생소하게 들리기도 해요.A. "석사 시절부터 파지(Phage) 기술을 활용한 연구를 해 왔습니다. 이를 기반으로 LG에서는 분자 클로닝(molecular clonning) 연구를 지속했고요. PD-1 면역항암제(항체) 시장은 이미 키트루다와 옵디보가 독점하고 있습니다. YBL-006만의 차별점이 있을까요?A. "전임상 과정에서 물성 시험을 했을 때, YBL-006의 물성이 월등히 좋았습니다. YBL-006은 키트루다 대비 5배 결합력이 좋았습니다. 또한 YBL-006의 경우 PD-1과 PD-L1 리간드가 결합하는 위치를 감싸는 것이 키트루다 대비 더 폭넓습니다. 이를 통해 높은 효능을 기대해 볼 수 있습니다."Q. 11명을 대상으로 한 1상에서는 모든 암종 환자 대상으로 진행했다고 하셨는데요, 확장 연구에서는 어느 암종을 타깃으로 할 계획인가요?A. 박영우 와이바이오로직스 대표 인터뷰코스닥 상장 앞둔 항체신약개발 기업1000억종 항체 라이브러리 세계 최고 수준T-세포 이중항체 플랫폼은 암젠 기술력 뛰어넘어항체 연구 20년 한 박 대표 필두로 글로벌 도약 나서 “현재 글로벌 시장에서 주목받고 있는 CAR-T(키메라 항원 수용체 T세포) 치료제와 ADC(항체약물접합체) 치료제 등은 모두 항체와 관련이 깊다. 결국 앞으로 신약개발에 있어서 항체가 가장 중요하다는 뜻이다. 우리는 세계 최고 수준의 항체 라이브러리 ‘Ymax®-ABL’을 구축했고, 이중항체 신규 플랫폼을 개발해 수 많은 기업의 관심을 받고 있다. 항체 플랫폼 기술 기반 글로벌 플레이어로 도약할 것이다.”26일 와이바이오로직스 대전 본사에서 이데일리와 만난 박영우 대표는 항체신약 전문기업으로서 글로벌 플레이어 도약에 강한 자신감을 내비쳤다. 2007년 항체신약개발 기업 와이바이오로직스를 설립한 박 대표는 서울대 미생물학 석사, 워싱턴대 박사 과정 후 LG화학(전 LG생명과학)과 한국생명공학연구원에서 항체 연구만 약 20년간 한 항체 전문가로 손꼽힌다.제약·바이오 업계는 물론 투자업계(IB)에서는 공통적으로 와이바이오로직스를 국내 최고 항체 기업이자 글로벌 수준의 기술력을 보유한 기업으로 평가하고 있다. 박 대표도 강한 자신감을 피력했다. 그는 “항체는 유전자를 통해 단백질을 만든다. 항체가 많으면 많을수록 좋은 후보물질을 찾을 수 있는 확률이 높아진다. 우리는 20년간의 항체 연구로 쌓은 기술력과 노하우로 세계 최고 수준인 1000억종의 항체를 발굴해 라이브러리화했다”며 “1000억종의 항체 중 약물 가능성이 높은 항체를 발굴해 신약을 개발할 수 있다. 예를 들어 코로나19 바이러스에 대한 중화항체 반응이 있는 항체를 골라 약물 최적화 과정을 거쳐 치료제나 백신으로 만들 수 있다”고 설명했다. 현재 상용화된 항체 기반 신약은 약 100여개 정도다. 대표적인 의약품으로는 자가면역질환 치료제인 휴미라(애브비)로 2020년 기준 연 매출이 20조원을 넘어섰다. 면역항암제로 주목받고 있는 키트루다(머크)와 옵디보(BMS) 등도 항체 기반 치료제다. 특히 글로벌 조사 기관 GBI리서치 보고서에 따르면 글로벌 면역항암제 시장규모는 매년 약 24% 증가해, 2022년에는 758억 달러(약 85조원)에 이를 것으로 전망된다.박 대표는 새로운 T-세포 이중항체 기술로 이들을 뛰어넘는 항암제를 개발하겠다고 강조했다. 그 중심에는 엘리스 플랫폼 기술이 있다. 엘리스 플랫폼 기술은 글로벌 시장에서 기적의 항암제로 떠오른 CAR-T 치료제와 1세대 항체 치료제를 대체할 것으로 평가받는다. 박 대표는 “CAR-T는 면역관문억제제의 낮은 반응과 한계를 극복해 우수한 약효를 입증했다”며 “하지만 고가의 치료비와 제한적인 적응증으로 시장 미충족 수요가 발생하고 있다”고 말했다.특히 암젠이 개발한 T-세포 이중항체 플랫폼 바이트(BITE)는 가장 앞서 상용화돼 높은 반응률과 완치율로 CAR-T 대안으로 주목받고 있지만, 독성 및 제한적인 적응증과 낮은 생산성으로 다양한 개선이 요구되고 있다. 반면 엘리스 플랫폼 기반 차세대 T-세포 이중항체 기술은 와이바이오로직스만의 고유 구조 T-세포 이중항체로 CAR-T와 1세대 T-세포 이중항체 치료제의 기술적 한계를 극복할 수 있도록 설계됐다는 게 박 대표 설명이다. 그는 “바이트 개발자도 우리 기술을 보고 처음 보는 기술이라며 깜짝 놀랐을 정도”라고 강조했다. 와이바이오로직스는 항체 라이브러리와 엘리스 플랫폼 기술 효과를 톡톡히 보고 있다. 1000억종 이상의 항체를 보유해 신약개발의 다양성과 높은 면역원성을 확보했고, CAR-T와 1세대 항체 치료제를 뛰어넘는 효능을 확보했기 때문이다. 박 대표는 “우리와 ADC 신약후보물질을 개발한 레고켐바이오가 미국 피식스 온콜로지사와 기술수출(약 3238억원) 계약을 체결하면서 지난해 계약금을 지급받았다. 중국 3D메디슨과 프랑스 3대 제약사 피에르파브르에는 이중항체 면역항암제 물질을 각각 약 952억원, 약 800억원에 기술수출했다”며 “이 외 HK이노엔과 한올바이오파마 등과 항체 기반 신약을 공동개발 중이고, 이들 모두 우리가 보유한 항체와 기술력을 보고 먼저 러브콜한 케이스”라고 말했다. 특히 “우리가 많은 항체를 보유하고 있다보니 항체 관련 공동연구 문의가 많이 들어온다. 약 120~130여개 회사가 우리와 관계를 맺고 있고, 셀트리온부터 유한양행은 물론 벤처기업까지 다양하다”고 했다. 올해 코스닥 상장을 앞둔 와이바이오로직스의 중장기 전략에 대해 박 대표는 이중항체 기반 면역항암제 개발과 세 번째 신약 플랫폼 개발을 강조했다. 박 대표는 “현재 국내와 호주에서 임상 1상을 진행 중인 면역관문억제제 ‘YBL-006’을 임상 2상을 마치고 희귀의약품 허가를 받아 이른 시일 내에 상업화시킬 것이다. 또 암 특성을 활용해 암에 특이적으로 붙어 변신이 가능한 신규 플랫폼 ‘카멜레온’을 개발 중”이라며 “글로벌 시장에서 해당 플랫폼이 기술이전 될 정도로 관심이 높아 긍정적인 결과가 예상된다”고 밝혔다. 와이바이오, 삼성서울병원과 "PD-1 등 IO" 공동연구·개발
임상1상 진행중 PD-1 'YBL-006' 임상개발 전략 수립 및 신규 면역항암제 발굴 및 검증 와이바이오로직스(Y-Biologics)가 삼성서울병원과 항암면역(immuno-oncology, IO) 치료법의 공동연구개발 상호협력을 위해 양해각서(MOU)를 체결했다고 16일 밝혔다.이번 와이바이오로직스와 삼성서울병원 협약의 주요 내용은 2가지다. 첫째, 와이바이오로직스가 진행하는 PD-1 항체 ‘YBL-006’의 임상 개발 적응증과 바이오마커를 찾고 임상 프로토콜을 디자인하는 등의 임상 전략을 수립하는데 협력한다. 와이바이로직스는 지난해 YBL-006의 국내, 호주 임상1상을 승인받은 바 있다.박영우 와이바이오로직스 대표는 “국내 첫 PD-1 항체로 임상에 진입해 원활하게 진행되고 있으며, 올해 안에 임상1상이 끝날 것으로 예상한다. 이에 임상2상을 앞두고 더 많은 환자가 면역항암제에 혜택을 받게 하기 위해 협력하게 됐다”며 “실제 병원과 협력하면서 ‘국산 PD-1’ 항체에 대해 큰 미충족수요가 있다는 것을 느꼈다. 글로벌 빅파마가 PD-(L)1 항체를 개발하고 있는 상황에서도 중국은 자체 PD-(L)1 항체를 성공적으로 시판했으며, 역으로 빅파마에 기술이전한 사례가 있다”고 설명했다. 이에 와이바이로직스도 국산 PD-1 항체 시판을 위해 속도를 낸다.둘째, 후속 면역항암제 파이프라인을 발굴하고, 항암 치료 효과를 높이기 위한 임상과학 연구와 검증을 진행한다. 삼성서울병원이 선진화된 의료역량과 이를 바탕으로 축적해온 임상경험과 유전체 기반의 데이터베이스는 와이바이오로직스의 인간항체 항체 발굴·개발 기술과 시너지 효과를 낼 것으로 기대한다.이규성 삼성서울병원 미래의학연구원 연구부원장은 ”이번 협력을 통하여 양 기관의 경험과 노하우를 공유함으로써 지속적인 연구과제 진행을 통해 좋은 결과가 나오기를 기대한다”고 말했다.박 대표는 “우수한 약효에도 불구하고, 기존의 면역항암제가 가지는 한계는 여전히 우리가 풀어야 할 숙제”라며 “글로벌 수준의 임상역량을 갖춘 삼성서울병원과의 협력으로, 보다 많은 환자들이 혜택을 받을 수 있는 면역항암치료법의 개발에 힘쓸 것”이라고 설명했다.김성민 기자 sungmin.kim@bios.co.kr

Y-Biologics Publishes Paper on Bispecific Antibodies Based on ALiCE Platform
Publication of research findings on YBL-013 licensed out to 3D Medicines and on the ALiCE platform in the journal, Biomaterials Set to expand the pipelines and currently developing an upgraded version of ALiCE Y-Biologics announced that the research findings on its bispecific T cell engager technology called Antibody-Like Cell Engager (ALiCE) and PD-L1- and CD3-targeting bispecific antibody, YBL-013, to which the platform was applied, had recently published in Biomaterials. ALiCE is a bispecific antibody technology in which the Fc domain of the antibody is substituted with VH-VL, which recognizes a different antigen. It is about 140 kDa in size. Fab (VH-VL), the original bivalent antigen recognition site, was maintained, while VH-HL that recognizes another antigen was incorporated into the Fc domain. This has resulted in a novel bispecific antibody with a 2:1 design that recognizes two different antigens. As for the background of coming up with this design, the first-generation bispecific T cell engager (BiTE, approx. 55 kDa) developed by Amgen has a short drug half-life, thus requiring ongoing injections and has had toxicity issues in activating T cells and application to normal tissue. In BiTE, VH and VL are connected at a ratio of 1:1. Y-Biologics’ novel format is expected to have specificity for cancer tissues where there is high expression of cancer antigens compared to normal tissues, based on the characteristics of binding to cancer antigens and T cells in a 2:1 ratio. In the published paper, Y-Biologics presents its research findings that showed the ALiCE platform, and YBL-013 (ACE-05), to which the ALiCE platform in an in vitro cancer cell model and in an in vivo mouse model, had lower toxicity and better ability to kill cancer cells compared to the BiTE platform. It was also confirmed that tumor growth was inhibited when YBL-013 was administered in a mouse model with humanized non-small cell lung cancer (HCC827+human PBMC). In primates, the drug half-life was found to be higher than that of BiTE. In cynomolgus monkeys, for instance, the half-life of BiTE was 9.0 hours, whereas the half-life of YBL-013 was more than three times higher at 31 hours. Bum Chan Park, the Head of Research at Y-Biologics, said, “This paper confirmed that our proprietary ALiCE platform has excellent competitiveness in the field of CD3-based bispecific T cell engagers. In addition to YBL-013, Y-Biologics has been discovering and developing a number of novel drug pipelines based on the ALiCE platform, and there are plans to further expand the pipelines. [...] Furthermore, we are in the stage of completing the upgrading process for the ALiCE platform to extend the half-life in the bloodstream, and we will apply it to target antigens of various types of solid tumor.” Meanwhile, four patents have been registered for the ALiCE platform in the United States, and patent applications have been submitted in major countries such as Korea, Europe, China, and Japan. Y-Biologics concluded a licensing agreement with 3D Medicines for the exclusive rights to the PD-L1xCD3 bispecific antibody, YBL-013, in China. 바이오머티리얼스(Biomaterials)에 ALiCE 플랫폼과 올해 1월 中 3D메디슨 기술이전한 'YBL-013' 연구 결과 발표..."파이프라인 확대 및 ALiCE 업그레이드 버전 개발중" 와이바이오로직스가 T세포 engager 이중항체 기술 ‘ALiCE(Antibody-Like Cell Engager)’ 플랫폼과 이를 적용한 PD-L1, CD3 타깃 이중항체 ‘YBL-013’에 대한 연구 결과가 바이오머티리얼스(Biomaterials)에 최근 게재됐다고 5일 밝혔다.ALiCE는 와이바이오로직스가 자체 개발한 이중항체 기술로 항체의 Fc 도메인을, 다른 항원을 인지하는 VH-VL로 치환한 형태다. 크기는 약 ~140 kDa 수준이다. 항체가 원래 갖고 있던 항원을 인지하는 2개의 항원 인식부위 Fab(VH-VL)은 유지하면서, Fc 도메인에 다른 항원을 인지하는 하나의 VH-HL이 결합한 것이다. 결과적으로 ‘2:1’ 디자인으로 2개의 항원을 인지하는 새로운 이중항체다.이러한 디자인을 고안한 배경으로 암젠의 1세대 T세포 engager 이중항체 ‘BiTE(Bispecific T-cell Engager, ~55kDa)’는 약물 반감기가 짧아 계속적인 주입이 필요하며, T세포를 활성화시키고 정상조직에 작용하는 등에 따른 독성 이슈가 나타났다. TME 등 초점맞춘 신규 면역항암제 타깃 최대 15개 대한 공동연구 및 기술이전...타깃당 최대 6000만유로 최대 9억유로 규모 와이바오로직스(Y-biologics)가 프랑스 3대 제약사인 피에르파브르 그룹(Pierre Fabre group)과 신규 면역항암제 공동연구 파트너십 및 옵션 라이선스 계약을 체결했다고 16일 밝혔다.앞서 지난해 11월 두 회사가 전략적 파트너십에 대한 의향서(LOI)를 체결한지 3달만에 최종 파트너십 계약이라는 점에서 의미가 있다. 피에르파브르는 2년전부터 면역항암제 파이프라인 확장을 위해 와이바이오로직스에 공동연구를 제의했으며, 이번 파트너십으로 양사의 공동연구가 탄력을 받을 것으로 기대된다.이번 공동연구 계약기간은 3년이며, 추후 2년의 계약 연장이 가능하다. 와이바이로직스는 매년 3개의 타깃을 연구하여 계약기간 동안 최대 15개의 타깃을 공동으로 연구 및 기술이전하게 된다.피에르파브르는 두 회사가 공동연구로 발굴한 선도(lead) 항체에 대한 라이선스 옵션을 갖게 되며, 타깃당 선급금, 개발 단계별 마일스톤, 판매 마일스톤을 모두 합쳐 최대 6000만 유로의 기술료를 지급받을 수 있다. 향후 제품화에 따른 로얄티는 별도다. 최대 15개 타깃이라는 것을 고려하면 계약 규모는 최대 9억유로에 이른다.와이바이오로직스는 항체의 전임상, 임상 개발에 참여할 수 있는 옵트인(opt-in) 옵션를 확보해 향후 피에르파브르와 항체 후보물과 공동연구를 이어나갈 수 있다.이번 공동연구에서 신규 타깃 면역항암제 약물을 개발할 계획이다. 와이바이오로직스는 자체 인간항체 라이브러리 Ymax®-ABL로부터 피에르파브르가 선정한 주요 항암 타깃에 대한 항체를 발굴하며, 피에르파브르는 생-줄리앙-앙-제네부아에 위치한 자사 면역센터(CIPF)에서 종양면역 전문지식을 제공 및 툴루즈 R&D센터에서 신약개발 업무를 담당한다.특히 이번 공동연구는 종양이 면역 반응을 방해하는데 중요한 역할을 하는 종양미세환경(TME) 세포 구성 요소를 주요 항원 타깃해, 고형암 환자의 면역체계를 회복하고 항암반응을 높이는 새로운 면역항암 요법을 제공하겠다는 목표다.박영우 와이바이오로직스 대표는 “피에르파브르와 같은 항암제 분야의 리더가 우리의 항체발굴 플랫폼을 인정하고, 한국 바이오 기업의 우수한 기술이 글로벌로 진출하게 되어 큰 자부심을 느낀다"고 말했다.또한 박 대표는 "특히 양사가 공동 연구하는 항체는 면역항암제가 잘 듣지 않는 원인인 고형암의 종양미세환경을 면역세포 친화적인 환경으로 바꿀 수 있는 타깃에 집중해, 자체 보유한 면역항암제 신약 파이프라인과의 시너지도 기대한다"며 "우리와 피에르파브르가 공동 개발한 후보 항체들이 전임상 및 임상 개발로 이어질 수 있기를 바란다”고 말했다.쟝-룩 로윈스키 피에르 파브르 의약부분 대표는 “면역항암제 연구는 피에르파브르의 최우선과제이며 이 분야에서 와이바이오로직스와 협업하게 된 것을 매우 기쁘게 생각한다. 첫 번째 종양 타깃에 대한 유망한 결과와 탄탄한 파트너십을 통해 양사는 곧 더 큰 종양 타깃 패널에서 추가 항체를 시험할 예정이다”라고 밝혔다.한편 와이바이오로직스의 Ymax®-ABL 플랫폼 기술은 인간 B세포의 cDNA로부터 유래된 완전 인간 항체 라이브러리로, 1000억 개 이상의 다양성을 보유하고 있다고 설명했다.김성민 기자 sungmin.kim@bios.co.kr

PD-L1xCD3 이중항체 'YBL-013'의 화학, 제조 및 품질관리(CMC)와 비임상, 임상시료 생산...와이바이오와 파트너사 3D메디슨 임상개발에 사용할 예정 와이바이오로직스(Y Biologics)가 지난달 CD3 기반의 이중항체 면역항암제 ‘YBL-013’의 비임상 개발 및 임상시료 생산을 위해 글로벌 CDMO(위탁개발생산기관) 업체인 우시바이오로직스(WuXi Biologics)와 계약을 체결했다고 3일 밝혔다.YBL-013은 와이바이오로직스가 독자적으로 개발한 T세포 이중항체 플랫폼인 ‘ALiCE’를 이용해 제작한 PD-L1 타깃 CD3 이중항체다. 지난해 12월 3D메디슨(3D Medicines)에 중국지역 전용실시권에 대한 라이선스아웃 계약을 체결한 바 있다. 와이바이오로직스는 YBL-013의 중국 이외 국내 및 글로벌 사업화에 대한 권리를 보유하고 있다.이번 위탁개발 계약은 YBL-013의 화학, 제조 및 품질관리(CMC)와 비임상, 임상시료 생산을 위한 것으로, 와이바이오로직스와 3D 메디슨이 공동으로 개발비를 분담하는 3자 계약이다.이를 기반으로 향후 와이바이오로직스는 비임상과 중국을 제외한 글로벌 지역의 임상시험을 진행하며, 3D메디슨은 YBL-013의 중국지역(대만, 홍콩, 마카오 포함) 사업화에 사용할 계획이다.박영우 와이바이오로직스 대표는 “최근 항체 및 바이오의약품에 대한 CMC의 중요성이 높아지고 있다"며 "특히 YBL-013과 같은 이중항체는 더욱 신중을 기하여 우수한 CMC를 유지하는 것이 중요하다"고 설명했다.박 대표는 "우시바이오로직스는 이중항체에 높은 이해도와 다양한 CMC 경험도 보유한 글로벌 톱 CDMO 업체이며, 사업 파트너인 3D Medicines와의 업무 효율도 고려한 전략적인 선택”이라며 “중국을 시작으로 YBL-013에 대한 글로벌 사업화에 속도를 내겠다”고 말했다ALiCE는 미국에서 특허 등록된 와이바이오로직스의 원천 기술로 2:1 형태의 독특한 구조를 가진 CD3 항체 기반 이중항체 플랫폼이다. 이러한 약물 디자인을 통해 암세포 항원에 더욱 강한 결합력을 가져, 암세포 주변의 T세포를 활성화시킬 수 있다고 회사측은 설명했다. 이에 따라 CD3 이중항체의 약물 독성을 낮출 것으로 기대한다.김성민 기자 sungmin.kim@bios.co.kr

Y-Biologics Passes Technical Evaluation for IPO on KOSDAQ
Set to file an application for preliminary review for listing on KOSDAQ in the first half of this year Y-Biologics passed the technical evaluation for IPO on the KOSDAQ under the exceptional technology scheme.. The company announced that it had passed the review grades of A and A from two evaluation agencies in the technical evaluation to be listed on the KOSDAQ under the exceptional technology scheme. In order for a company to be listed on the stock exchange under the conditions of having an exceptional technology, it needs to obtain grades of at least A and BBB from two evaluation agencies designated by the Korea Exchange. “Through this technical evaluation, the excellence of the immuno-oncology technology and bispecific T cell engager technology that our company has built has been recognized,” said Y-Biologics CEO Young Woo Park who added, “Based on this, we will swiftly proceed with the IPO procedure to be listed on KOSDAQ.” Y-Biologics plans to submit a preliminary review application to be listed on the KOSDAQ in the first half of this year. Korea Investment & Securities is the underwriter. Y-Biologics is a biotech company specializing in antibody drug R&D that was founded by Dr. Young Woo Park, who had led the research for antibody library and new antibody drug development at LG Life Sciences and Korea Research Institute of Bioscience and Biotechnology. The company has developed a human antibody library for discovering new antibodies and a new format of CD3-based bispecific antibodies suitable for bispecific T cell engager technology. Y-Biologics has also licensed out its technology recently. The company jointly developed an antibody-drug conjugate (ADC) as a new drug candidate that targets DLK-1 with LegoChem Biosciences and licensed it out to Pyxis Oncology for a total of USD 294 million. It has also licensed out a CD3xPD-L1 bispecific T cell engager to 3D Medicines, a Chinese biopharmaceutical company, for a total of USD 85 million, which includes milestone fees for clinical development approval, commercialization, and sales in China.
